VIRTUAL SERUM PREPARATION FOR UNEXPECTED ANTIBODY SCREENING IN MEDIA OF NORMAL SALINE SOLUTION, LOW IONIC STRENGTH SALINE AND PAPAIN ENZYME

Abstract

Objective: To prepare virtual serum that has a yellow, light yellow and turbid yellow color, similar to the color of serum obtained from human blood for use in teaching the laboratory of transfusion science subject, Faculty of Medical Technology, Rangsit University in the topic of antibody screening for unexpected antibody detection.

Methods: Virtual serum that has yellow and light yellow from previous study were prepared. This study has developed the preparation of serum that is turbid yellow color. The naked eyes reviewed the appearance of the virtual serum. Optical density and specific gravity of virtual serum were measured and compared using the Kruskal-Wallis test statistics. Antibody screening that gave negative and positive results was performed using convention tube test (CTT) in media of normal saline solution, low ionic strength saline and papain enzyme.

Results: Virtual serum with turbid yellow color was more turbid than the yellow and light yellow.

The optical density and specific gravity of  three types of virtual serum were statistically significant different (p < 0 . 050). For the specific gravity, it was found that the light yellow virtual serum had statistically significant different from the yellow and turbid yellow virtual serums ​​(p < 0.05). The results of the antibody screening  in all media showed that the three types of virtual serum without adding anti-D (IgG) gave true negative and did not produce false positive. The virtual serum with added anti-D (IgG) gave true positive and did not produce false negative results.

Conclusion: The yellow, light yellow, and tubbid yellow virtual serums gave different yellow and turbidity. The preparation of several samples with physical appearance in terms of color and turbidity that were similar to human serums for use in teaching the laboratory of antibody screening in media of normal saline solution, low ionic strength saline, and papain enzyme effectively and efficiently.
