การศึกษาสมบัติและฤทธิ์ต้านเชื้อราของไมโครอิมัลชันน้ำมันกานพลู บรรจุฟลูโคนาโซล

บทคัดย่อ

การนำส่งยาเฉพาะที่จัดเป็นวิธีการที่มีประสิทธิภาพในการรักษาการติดเชื้อราที่ผิวหนัง เนื่องจากช่วยลดความเสี่ยงของผลข้างเคียงต่อระบบต่างๆ ในร่างกาย อย่างไรก็ตาม การออกแบบตำรับยาที่ประสบความสำเร็จนั้นจำเป็นต้องเพิ่มประสิทธิภาพการซึมผ่านของยาเข้าสู่ผิวหนังให้เพียงพอ งานวิจัยนี้มีจุดมุ่งหมายเพื่อพัฒนาไมโครอิมัลชันน้ำมันกานพลูบรรจุฟลูโคนาโซลร้อยละ 2 โดยน้ำหนัก คัดเลือกไมโครอิมัลชัน 4 ตำรับจากพื้นที่ไมโครอิมัลชันที่ได้จากการสร้างแผนภาพวัฏภาคไตรภาคเทียม ซึ่งประกอบด้วยน้ำมันกานพลู ของผสมระหว่างนํ้ากับโพรไพลีน ไกลคอล (propylene glycol) ในอัตราส่วน 1:1 และทวีน 80 (Tween 80) ประเมินตำรับในด้านสมบัติรวมถึงฤทธิ์ต้านเชื้อรากลุ่มแคนดิดา (Candida species) 4 สายพันธุ์ ได้แก่ C. albicans, C. krusei, C. lusitaniae และ C. tropicalis ผลการทดลองพบว่าตำรับที่เตรียมได้ทั้งหมดเป็นไมโครอิมัลชันชนิดน้ำมันในน้ำ ลักษณะเป็นของเหลวใสสีเหลือง ค่าความเป็นกรด-ด่างและความหนืดอยู่ในช่วงที่ยอมรับได้สำหรับการใช้ทาภายนอก ไมโครอิมัลชันน้ำมันกานพลูทั้งหมดแสดงฤทธิ์ต้านเชื้อราสายพันธุ์ที่ทดสอบ ซึ่งสามารถอธิบายได้ว่าเป็นผลของยูจีนอลซึ่งเป็นส่วนประกอบหลักของน้ำมันกานพลู เส้นผ่านศูนย์กลางของพื้นที่ยับยั้งเชื้อมีขนาดในช่วง 23.0-30.3 มิลลิเมตรสำหรับไมโครอิมัลชันบรรจุฟลูโคนาโซลซึ่งใหญ่กว่าอย่างมีนัยสำคัญเมื่อเทียบกับตำรับเปล่า งานวิจัยนี้แสดงให้เห็นว่าไมโครอิมัลชันน้ำมันกานพลูบรรจุฟลูโคนาโซลที่พัฒนาขึ้นมีแนวโน้มในการใช้เป็นตำรับเฉพาะที่เพื่อต้านเชื้อราที่ผิวหนังได้ และควรนำไปศึกษาเพิ่มเติม ตำรับที่น่าสนใจมากที่สุดสำหรับการศึกษาเพิ่มเติม คือ FLZ-ME-1 ซึ่งประกอบด้วยฟลูโคนาโซล น้ำมันกานพลู ของผสมระหว่างนํ้ากับโพรไพลีน ไกลคอล ในอัตราส่วน 1:1 และทวีน 80 ร้อยละ 2.0, 9.8, 49.0 และ 39.2 โดยน้ำหนัก ตามลำดับ
