บทคัดย่อ

การวิจัยนี้มีวัตถุประสงค์เพื่อเพิ่มประสิทธิภาพการแยกและเพาะเลี้ยงเซลล์ไฟโบรบลาสต์ และเซลล์เยื่อบุผิวปฐมภูมิจากเหงือกมนุษย์ให้ได้เซลล์ที่มีความบริสุทธิ์สูง และปราศจากการปนเปื้อนข้ามชนิดของเซลล์ภายหลังการถ่ายเลี้ยงเซลล์ในครั้งแรก (P1) โดยเก็บตัวอย่างชิ้นเนื้อเหงือกจากอาสาสมัครสุขภาพดี จำนวน 5 ราย ที่เข้ารับการรักษา ณ คลินิกศัลยกรรมปริทันต์ คณะทันตแพทยศาสตร์ มหาวิทยาลัยขอนแก่น จากนั้นเพาะเลี้ยงด้วยวิธี Direct explant และแยกเซลล์ด้วยทริปซิน-อีดีทีเอความเข้มข้นร้อยละ 0.25 และ 0.05 ร่วมกับอาหารเลี้ยงเซลล์ที่มีความจำเพาะต่อชนิดเซลล์ พร้อมทั้งประเมินประสิทธิภาพของวิธีการแยกและเพาะเลี้ยงโดยการตรวจสอบลักษณะความสอดคล้องทางสัณฐานวิทยาเปรียบเทียบกับเซลล์มาตรฐาน


ผลการวิจัยพบว่าภายหลังการถ่ายเลี้ยงเซลล์ครั้งแรกสามารถเพาะเลี้ยงเซลล์ปฐมภูมิจากเหงือกได้สำเร็จในทุกตัวอย่าง (ร้อยละ 100) โดยกลุ่มตัวอย่างจำนวน 4 ใน 5 (ร้อยละ 80) ปราศจากการปนเปื้อนข้ามชนิดของเซลล์ อีกทั้งยังมีลักษณะทางสัณฐานวิทยาที่สอดคล้องกับเซลล์มาตรฐาน และมีความบริสุทธิ์สูงกว่าร้อยละ 95 ขณะที่พบการปนเปื้อนข้ามชนิดเพียง 1 ตัวอย่าง (ร้อยละ 20) สรุปได้ว่า วิธีการแยกเซลล์แบบผสมผสานที่พัฒนาขึ้นสามารถเพิ่มประสิทธิภาพการแยกเซลล์ปฐมภูมิให้มีความบริสุทธิ์สูง ช่วยลดต้นทุนและความสูญเสียจากการเพาะเลี้ยงเซลล์ปนเปื้อน จึงมีความเหมาะสมสำหรับนำไปประยุกต์ใช้เป็นมาตรฐานการปฏิบัติงานประจำของห้องปฏิบัติการต่อไป
